To address these questions, we will establish at least one ELN certified lab per country being able to serve as national reference lab. An exchange program for scientists and technicians will allow spread of quality controlled, standardized PCR methodologies all over Europe. Regular control rounds using patient material (lysates of CML cells in normal cells) will ensure high quality reliable results in the monitoring of CML patients in all countries. We will analyse the spectrum and frequency of mutations after imatinib and second generation tyrosine kinase inhibitors and determine the therapeutic implications. Further, we will analyze the frequency of KD mutations in a cohort of imatinib-naive patients with CML in chronic phase, accelerated phase, or blast crisis. We will use a panel of newly developed fluorescencebased allele-specific PCR assays for high-sensitivity detection of the most common mutations as well as conventional sequencing of PCR products amplified from cDNA and new recently described techniques like the "colony assay". The detection of KD mutants will then be correlated with baseline variables and outcome.
